Stereoselective cyclic peptide assisted DART-MS for rapidly screening environmental disruptors of dopamine receptors

The emerging binding-directed analysis (BDA) technique has successfully employed stereoselective oligopeptide probes to capture G protein-coupled receptor (GPCR)-active compounds from complex environmental matrices via electrospray ionization (ESI)-enabled complex formation. However, its integration with direct analysis in real time-mass spectrometry (DART-MS), a technique characterized by fundamentally different ionization mechanisms and exceptionally high analytical throughput, remains unexplored. This study evaluates a DART-MS-based BDA workflow for rapid, chromatography-free screening of GPCR ligands using ten model dopamine (DA) receptor ligands and a dopamine-selective cyclic peptide probe (CPP). The workflow required less than 1 min per measurement. Notably, DART-MS even achieved an average CPP-ligand complexation efficiency substantially higher than that obtained in ESI-MS (20% vs 4%), facilitating reliable identification of CPP-ligand complex ions from redundant mass spectra. Moreover, most CPP-ligand gas-phase complexes formed in DART-MS and ESI-MS showed comparable collision stability. Only the CPP-sulpiride and CPP-pramipexole complexes showed stability reduction by 85% and 61% in DART-MS compared to ESI-MS. These findings suggest that DART desorption/ionization conditions may promote more efficient formation and/or preservation of CPP-ligand complex ions. The applicability of the DART-MS-based BDA workflow was further validated through analysis of surface water samples. Although the spiked model ligand perphenazine (L1) was not captured by the CPP, the rubber additive 1,3-diphenylguanidine was successfully identified, a DA antagonist formerly also discovered by the ESI-MS-based BDA workflow. Collectively, this study establishes DART-MS-based BDA as a highly-efficient screening tool for future large-scale surveillance of GPCR-disrupting chemicals in the environment.

Pramipexole promotes CD8+ regulatory T cell-dependent neuroprotection in Parkinson’s disease patients

BACKGROUND:
Parkinson's disease (PD) is caused by the death of dopaminergic neurons. Since immune system cells express dopamine receptors, dopaminergic agonists may modify their function. However, the effects of dopaminergic agonists on immunoregulation and the specific dopamine receptors involved are unknown.
OBJECTIVE:
This study will examine the effects of pramipexole treatment on dopamine receptor expression and immunosuppressive activity of CD8+ Tregs, as well as its neuroprotective effects.
MATERIAL AND METHODS:
Dopamine receptor expression was determined in patients treated with pramipexole and in control subjects. PBMCs from controls were stimulated with anti-CD3/CD28 alone or with 2-200 ng/mL of pramipexole for 24 h. CD8+ T cell immunosuppressive capacity in both groups was evaluated using flow cytometry. CD8+ Tregs were pretreated with pramipexole and co-cultured with dopaminergic neurons previously exposed to 6-OHDA. Neuroprotective capacity was defined by the survival of Tyrosine Hydroxylase-positive neurons, as determined by immunofluorescence.
RESULTS:
Pramipexole treatment increased the proportion of CD8+ T cells expressing D2 receptors, and in vitro studies showed similar results. The suppressive function of CD8+ Tregs was similar in patients treated with pramipexole and control subjects. This suggests that dopaminergic treatment compensates for the reduction in the number of CD8+ Tregs. Pramipexole (20 and 200 ng/mL) increased the suppressive function of CD8+ Tregs. CD8+ Tregs stimulated with 2 or 20 ng/mL of pramipexole exhibited a neuroprotective effect on dopaminergic neurons challenged with 6-OHDA.
CONCLUSION:
Pramipexole increased the percentage of CD8+ T cells expressing the D2 receptor on their cell membranes. Treatment of CD8+ Tregs improved their immunosuppressive function and had a neuroprotective effect. Therefore, in addition to its previously reported direct effects on neurons and glial cells, treatment with pramipexole may also exert immunomodulatory effects on CD8+ regulatory T cells.

Pramipexole mitigates rotenone-evoked hepatic injury by regulating inflammatory, antioxidant, apoptosis and NLRP3-mediated pyroptosis

The environmental chemical rotenone, which is employed as a natural insecticide, is hazardous to many organs. Thus, this study aimed to elucidate the protective efficacy of an oral administration of pramipexole (PPX) at a dose of 0.5 mg/kg/d against rotenone-induced hepatocellular damage for 60 days. Rotenone administration caused significant hepatic dysfunction, as evidenced by elevated serum AST, ALT, GGT, and bilirubin levels, along with reduced total protein and albumin, indicating impaired liver function. PPX co-administration markedly attenuated these changes, restoring liver enzyme activities and protein levels toward normal values but without a significant effect on albumin level. Additionally, rotenone-induced dyslipidemia was evident through increased total cholesterol, triglycerides, LDL, and VLDL, and decreased HDL levels. PPX significantly improved these lipid disturbances, demonstrating its hypolipidemic potential. Oxidative stress markers revealed that rotenone elevated MDA and GSSG while decreasing SOD and GSH levels. PPX pretreatment notably restored antioxidant balance by reducing lipid peroxidation and enhancing GSH-dependent defenses. At the molecular level, rotenone upregulated pyroptotic (NLRP3, ASC, Caspase-1, GSDMD, IL-18) and inflammatory (TNF-α, IL-1β, MCP-1, MPO) markers while downregulating hepatic miRNA-143-3p expression. PPX co-treatment significantly reversed these alterations, demonstrating anti-inflammatory and anti-pyroptotic efficacy but without any significant effect on miRNA-143-3p. Histopathological and immunohistochemical analyses further confirmed PPX's hepatoprotective role, showing near-normal hepatic structure, reduced necrosis, and suppressed expression of apoptotic (caspase-3, BAX) and macrophage activation (CD68) markers. Overall, PPX pretreatment effectively ameliorated rotenone-induced hepatotoxicity through antioxidant, anti-inflammatory, and anti-apoptotic mechanisms, suggesting its potential therapeutic value in preventing toxin-induced liver injury.
